Fighting bacteria with mucus

Slimy layers of bacterial growth, known as biofilms, pose a significant hazard in industrial and medical settings. Once established, biofilms are very difficult to remove, and a great deal of research has gone into figuring out how to prevent and eradicate them.

Results from a recent MIT study suggest a possible new source of protection against biofilm formation: polymers found in mucus. The MIT biological engineers found that these polymers, known as mucins, can trap bacteria and prevent them from clumping together on a surface, rendering them harmless.

"Mucus is a material that has developed over millions of years of evolution to manage our interactions with the microbial world. I'm sure we can find inspiration from it for new strategies to help prevent infections and bacterial colonization," says Katharina Ribbeck, the Eugene Bell Career Development Assistant Professor of Biological Engineering and senior author of the paper, which appears in the Nov. 8 online edition of the journal Current Biology.

Mucin coatings may help prevent biofilm formation on medical devices and could also find applications in personal hygiene: Incorporating them into products such as toothpaste or mouthwash may supplement the body's own defenses, especially in people whose natural mucus has been depleted, Ribbeck says.

Lead authors of the Current Biology paper are former MIT postdoc Marina Caldara and Ronn Friedlander, a graduate student in the Harvard-MIT Division of Health Sciences and Technology. Other authors are Nicole Kavanaugh, an MIT graduate student in biology; Joanna Aizenberg, a professor of materials science at Harvard University; and Kevin Foster, a professor of evolutionary biology at the University of Oxford.

How to stop bacteria from teaming up

Mucus normally lines most of the wet surfaces of the body, including the respiratory and digestive tracts. "The textbook view of mucus is that it forms a barrier to infection, but it's not at all clear how it does so," Ribbeck says.

To investigate that question, Ribbeck and her colleagues observed the behavior of Pseudomonas aeruginosa bacteria in a growth medium that contained soluble purified mucins ? long proteins with many sugar molecules attached.

For bacteria to effectively penetrate the mucus layer and infect the tissues below, they need to form clusters that can adhere to the tissue surface. Clumps of bacteria are much more difficult for the immune system to clear, because immune cells are specialized to attack individual bacterial cells.

"In general, you want to have bacteria around, you just don't want them to team up," Ribbeck says. "You want to them to be mixed with many other bacteria that are good for you. You don't want a single species to take over, because then they may overgrow the system."

In the new study, the researchers found that mucins block bacterial cluster formation by preventing them from adhering, which is necessary for them to clump together. When bacteria stay motile, they end up suspended in a gooey mix and can do less harm.

"The mucins have the ability to suppress virulence by keeping the cells separate. It's like keeping your kids in separate rooms, so they will stay out of trouble," Ribbeck says.

However, bacteria are sometimes able to break through this defense system and cause infections. This can be accelerated by reductions in mucus due to aging, dehydration or chemotherapy, Ribbeck says. Or it may be that the mucus does not get replaced often enough, as happens in the mucus-clogged lungs of cystic fibrosis patients.

'Managing microbial behavior'

One advantage of using mucins as antimicrobial coatings is that the substance disarms pathogenic bacteria without killing them. This makes it less likely that bacteria could evolve resistance to mucins, as they do to antibiotic drugs. It would also spare the beneficial bacteria that live on mucus membranes.

"This is a nice mechanism where you just suppress the virulence traits without killing the bacteria," Ribbeck says. "It's nature's way of managing microbial behavioral in a way that could be useful to take advantage of."

Her lab is now investigating exactly how mucins prevent bacteria from losing their motility, and also how they block infection by nonmotile bacteria. Mucins seem to have wide-ranging antimicrobial properties: Ribbeck has previously shown that they can trap viruses and keep them from infecting cells, and she is now studying mucin interactions with other pathogenic organisms, such as yeasts.

BARBOURVILLE, Ky. (WKYT) - Beverly Foley has so many fond memories of November 9th. It was the day she was born?and the day 27 years ago she gave birth to her son?.Jonathan.

?I miss him terribly. I got this morning..each day gets worse..instead of better,? said the missing man?s mother Friday.

Since the national guardsman was serving his country in Iraq last year?.and involved in other activties before?this would have been the first birthday in several years that the two would have spent together.

?I?m hoping something today?with it being his birthday..will bring it to closure,? said Foley.

Foley was last seen Tuesday heading to Lee?s Food Mart in Barbourville. His truck was found covered in mud several miles away?near a factory close by Knox County Hospital.

His family says what was found inside that truck was very unusual.

?The seat was kind of scooted up. The driver?s seat. He?s a very tall person,? said Foley.

The Foleys say police told them they were planning to search the truck and gather evidence on Friday. Police were also said to be gathering surveillance video from the area where the truck was found. His family meanwhile is hoping for the best but fearing the worst.

?I?m feeling lost without him. I?m afraid he won?t be coming home. I hope he will,? said Charles Foley, Jr., the missing man?s father.

Tentacled snakes born at National Zoo

Eight tentacled snakes were recently born in captivity (Wikicommons)Indiana Jones and Samuel L. Jackson may need to start their own group therapy session: Eight tentacled snakes were recently hatched at the Smithsonian's National Zoo.

These babies belong to an aquatic species of snake native to Southeast Asia, formally known as Erpeton tentaculatus, according to the Mother Nature Network.

Scientists say it is the only known species of snakes to have tentacles, which are used to sense vibrations from fish swimming by. The snakes are relatively small, usually growing to lengths of about 20 to 35 inches.

The tentacled snake spends almost all of its life hiding in murky waters, and scientists say its venomous fangs are not a threat to humans.

Beyond its unusual sensory devices, the tentacled snake has some other impressive evolutionary traits?as well, including the ability to predict where a startled fish will maneuver, allowing the tentacled snake to literally guide the prey into its open mouth. A 2009 report from Science Blogs explains in detail how tentacled snakes use their sharp senses and incredible speed to trap their prey.

"I haven't been able to find reports of any other predators that exhibit a similar ability to influence and predict the future behavior of their prey," Vanderbilt University professor of biological sciences Kenneth Catania said in a statement.

And in unsettling development for those suffering from ophiophobia, or the fear of snakes, the tentacled snakes also reportedly grow very quickly.

"Within a few hours of being born, the snakes were already acting like adults," Matt Evans, a keeper at the Smithsonian's Reptile Discovery Center, said in a statement. "Instincts took over, and they were hunting. We don't know much about this cryptic species, but we're already learning so much just watching them grow."

For those living in or visiting the Washington, D.C., metro area, four of the baby snakes are currently on display at the National Zoo's Reptile Discovery Center.

On Twitter, pope to get different type of followers

VATICAN CITY (Reuters) - Pope Benedict already has 1.2 billion "followers" in the standard sense of the word but he soon will have another type when he enters what for any 85 year old is the brave new world of Twitter.

Vatican officials say the pontiff, who is known not to love computers and still writes most of his speeches by hand, will have his own handle by the end of the year.

"It will be an officially verified channel," said a Vatican official.

Primarily the tweets will come from the contents of his weekly general audience, Sunday blessings and homilies on major Church holidays. They will also include reaction to major world events, such as natural disasters.

The leader of the world's 1.2 billion or so Roman Catholics will not, of course, write the tweets himself, but he will sign off on them before they are sent in his name.

But even divine intervention might not help squeeze the gist of a papal encyclical, which can run to more than 140 pages, into 140 characters.

Those tweets will probably be limited to a link to a url with the entire document.

The papal handle has not yet been disclosed but it is widely expected to be @BenedictusPPXVI, his name and title in Latin.

The pope has given a qualified blessing to social networking.

In a document issued last year, he said the possibilities of new media and social networks offered "a great opportunity", but warned of the risks of depersonalisation, alienation, self-indulgence, and the dangers of having more virtual friends than real ones.

In 2009, a new Vatican website, www.pope2you.net, went live, offering an application called "The pope meets you on Facebook", and another allowing the faithful to see the pontiff's speeches and messages on their iPhones or iPods.

The Vatican famously got egg on its face in 2009 when it was forced to admit that, if it had surfed the web more, it might have known that a traditionalist bishop whose excommunication was lifted had for years been a Holocaust denier.

I doubt they will as the majority of funding comes from BPA. Also the returns are similar to what the Kalama and Lewis have experianced over the last several years, to Plant 350K smolts and not harvest 100 adults is a joke.

The Kalama had some great years when they were planting Carson upriver stock. But HSRG put a stop to that and returns have plumeted since not sure why they plant fish that dont return in significant numbers but they should focus on results instead of how many turds they release.

Not too many years ago I caught many springers in the Elochoman while summer steel fishing and was told these were Deep river strays, what were they doing 5 years ago that they are not doing now?

Stocking springers in Willapa Bay would be poor fisheries management. Sorry to be so direct, but spring Chinook need cold water all summer so they can spawn in late August or September. The Willapa Bay tribs do not flow off the Cascades (like the Cow, Lewis, and Kalama) so they get really warm during the summer. That would be a death trap for springers.

I agree with Slab on the Kalama River. Back in 2007, they got 7000 springers back to the rack. That was twice as many as they've ever seen. Plus the recreational fishing was outstanding that year. Off the charts. Since then, they've closed the season early every year because they're not getting any fish back. That happened in 2008, 2009, 2010, 2011, and again this past spring. In fact, this year the Kalama Rv hatchery did not make their egg take for springers, so they got some from the Cowlitz hatchery.

What's worse is that since 2007, Kalama Rv spring Chinook don't show up in any fishery - recreational, commercial or Tribal. So whatever they're doing isn't contributing to the fishery resources of the State of Washington. I'm hoping they change back to Carson stock, if they haven't already. I'm not sure springers were ever native on the Kalama Rv so I'm not sure why they're concerned about wild fish.

25+ years ago, Tom Pent, a WDFW person, and someone from the Chehalis tribe,and myself went to the holding ponds below the dam on the Skookumchuck. The purpose, was to see if those holding ponds would be OK to be used for Spring Chinook.

That was before the "merger" of Salmon and Wildlife departments. At some point in time, springers went by the wayside and the ponds were used for Coho for PS.

While you might be correct on the "more problems than solutions"....just seems that politics gets involved too much of the time in Region 6.

Of course you're correct.......the river that has the "best chance" got written off when Dick Stone was the head WDFW bio in Region 6.

Lot's of meetings, to much "spinning of wheels", fight, fight fight.....mini hatchery below Wynoochee Dam, then holding ponds......now all these years later.....nothing.......springers got "killed off", can't get Region 6 to want to do anything with mitigation monies. Last I heard,.........well what I heard...no fish were to be raised..........grrrrrrrr $2.4 million.... to be pissed away.

It seems like it would be very counter productive to add additional Springers to the Chehalis. Yes, it already has a native run as does the Newaukum and the Skookunchuck. I say it would not be a good use of resource money just due to the extensive netting by the QIN. Any more fish in the system would increase netting pressure and put even more pressure on the native steelhead that are in the system at the same time frame.

QIN pretty much net, 12/1 - 5 or 6/1, and have for many years. Nat's and late Silvers, pretty much gone on Wynoochee, Satsop, and Chehalis.

What is a "good use of resource money"......if you have any ideas.....let fish management, in Region 6, know.......they don't have a clue.

Today's Papers: Defense Spending, Health Insurance Ain't a Normal ...

A few pieces in the papers today on themes near and dear to the OTE heart.

First, while Gov. Romney?s proposed increase in defense spending became an important Obama talking point, the fact is that the military industrial complex that Eisenhower warned about remains alive and well, as discussed compellingly here.? One point the author makes really hit home for me from a few interactions I myself had in my White House days: the knee-jerk calculus on military spending by civilian politicians.

Obviously, Ike?s authority came from his military experience, but it?s rare, though thankfully increasingly less so, for civilian policy makers to question military spending.? It?s considered a stroke of boldness, if, like the President, you propose to slow its growth.

None of which is to say we should indiscriminately cut, but that, as I discuss at the end of this post, we should listen to people like Ike, who have the insider experience to make educated calls on what?s needed and what isn?t (follow the link to the Larry Korb piece, e.g.).

BTW, I recently cited Ed Luce?s book, Time to Start Thinking, about America?s current denial about many of our economic challenges.? He?s especially good on this one, talking to many defense experts who recognize the defense spending problem documented in the oped and how it is weakening the nation rather than strengthening it.

Next, a theme I haven?t visited for a while here is the old OTE saw that health care isn?t a normal market such that you?d expect competition to work the way Republican voucher plans presume it will.? Here?s a great example of a highly educated health care expert who herself has serious health needs trying to negotiate the system.

And if it?s this hard for her, imagine what it would be like for the average Joe and Jane.? For the life of me, I?ll never understand why ?Instead of guaranteed coverage under Medicare as its currently structured, we?ll let you shop for your insurance plan!? is a selling point.

Finally, here?s a piece amplifying the fact-abuse in this election cycle that struck many of us who?ve been around for a while as beyond the usual limits.? There?s a difference between bending and breaking facts, and I agree with the author here that the Romney team were terrible fact breakers.? More so than any campaign I?ve ever seen, they worked from a playbook that tried to take advantage of today?s ?truthiness? zeitgeist?the idea that making stuff up is fine if it?s part of your ?narrative? even if it?s belied by the facts.

Romney?s whole campaign can be usefully understood in this light: say what you will to win the severely conservatives hearts of Republican primary voters and then drop it all to capture the independents who will decide this thing.

And today we?ll learn if it worked.
